چکیده

Helicobacter pylon (H pyloon) infection is associated with hypo, normal, and hypersecretory disorders of the gastric mucosa. Pathophysiologicalpathways bywhichHpylori interacts with acid secretion are still unclear. The effects of H pylon on (14C) aminopyrine uptake by human parietal cells were examined as an indirect assay for acid secretion. Isolated oxyntic glands were stimulated with submaximal concentrations of histamine or carbachol and incubated with sonicates of different H pylon strains. Omeprazole and sonicates of Campylobacter jejuni served as positive and negative controls, respectively. Two of four H pylori strains reduced hydrochloric acid sequestration within the parietal cells significantly and in a dose dependent manner in up to 80%. Interaction with acid secretion may therefore constitute a factor contributing to a distinct pathogenicity of Hpylon strains. Accepted for publication 20 September 1993 Helicobacter pylon (H pylon) causes or is at least associated with different clinical and histological entities such as chronic gastritis, peptic ulcer disease, and even gastric carcinoma. Under these conditions acid secretion varies from hypo to hyperchlorhydria. Recently it has been shown that sonicates of H pylon strongly inhibit acid secretion by rabbit parietal cells.' Rabbits, however, are not natural hosts ofH pylon. In humans H pyloni infection may cause acute epidemic gastritis associated with hypochlorhydria.2-4 Whether hypochlorhydria occurs in every short term H pylon infection is as unknown as the influence of longterm H pylon infection on parietal cell function. We have examined the effects of sonicates ofdifferentHpylon strains on acid secretion from isolated human oxyntic glands, using '4C-aminopyrine ('4C-AP) uptake by the parietal cells. This well established assay indirectly reflects acid production.

Effect of N alpha-methyl-histamine on acid secretion in isolated cultured rabbit parietal cells: implications for Helicobacter pylori associated gastritis and gastric physiology.

BACKGROUND Helicobacter pylori has been shown to produce the unusual metabolite N alpha-methyl-histamine. This compound is known to be a potent agonist at inhibitory histamine H3 receptors. There is increasing evidence implicating this receptor in the control of gastric acid secretion but the mechanism for this remains to be clarified. AIMS To investigate the effect of N alpha-methyl-histamin...

Effect of purified lipopolysaccharides from strains of Helicobacter pylori and Helicobacter felis on acid secretion in mouse gastric glands in vitro.

As a bacterial product, Helicobacter pylori lipopolysaccharide (LPS) can originate in close proximity to parietal cells, but the role of this uniquely structured endotoxin on acid secretion has not been fully investigated and remains unclear. The purpose of this study was to test the direct effect of purified LPS (tested range, 0.1 to 100 microg/ml) from various strains of H. pylori and from on...

The effect of intragastric D-glucose on hyperacidity induced by histamine and carbachol in male rat

It has been shown that enhanced acid secretion can be suppressed in the rat by systemic administration of glucose. We have recently reported that enhancement of acid secretion as a result of pentagastrin was diminished by intragastric administration of D-glucose. Present study was designed to investigate whether intragastric D-glucose influences gastric acid secretion associated with carbachol ...

Effect of thyroid hormones on basal and stimulated gastric acid secretion due to histamine, carbachol, and pentagastrin in rat (an in vitro study)

Although thyroid hormones are known to influence acid secretion, but exact mechanisms are not fully understood. So, in this experimental study carbachol, histamine, and pentagastrin were used to stimulate acid secretion in isolated stomach of hypo- and hyperthyroid rats and data were compared with controls. Each group consisted of 8 N-Mari rats from both sexes weighing 246 ± 5 g. Hypo- and hype...

The role of D1/DA1 dopamine receptors on histamine-and carbachol-induced gastric acid secretion in male rat

Dopamine has an effective role on gastric acid secretion (GAS). The role of peripheral and central dopamine D1/DA1 receptors on stimulated GAS are unknown. The objective of the present study was to use SKF38393 and SCH23390 on stimulated GAS. For all gastric sampling, animals (Sprague Dawley rats) were anesthetized and a polyethylene tube was introduced into the stomach through esophagus and a ...
